Peter Parker (Andrew Garfield) cerca di trovare un equilibrio nella sua doppia vita da ragazzo innamorato di Gwen Stacy (Emma Stone) e Spider-Man. Avere a che con questo conflitto interiore e con le responsabilità che pesano sulle spalle di un supereroe non è facile. Ma una minaccia più grande è in agguato. Solo lui può proteggere i suoi concittadini dai nemici pericolosi che assediano New York. Con la comparsa di Electro (Jamie Foxx), Peter deve affrontare un nemico molto più potente di lui, e quando il suo vecchio amico Harry Osborn (Dane DeHaan) ritorna, Peter si rende conto che tutti i suoi nemici hanno una cosa in comune: la OsCorp.
